Proclinical is seeking an experienced principal scientist or associate director to join our motivated and dynamic AAV gene therapy-based research team. The successful applicant will be responsible for contributing to the discovery and development of AAV vectors for gene therapy across our therapeutic target pipeline. A main focus of this position is to establish and lead our internal AAV production platforms to support novel capsid discovery and early pipeline program development. The position requires extensive knowledge with hands-on experience working in molecular and cell biology, virology, protein expression, AAV production and purification.
Job Responsibilities:
- Conduct viral vector design, capsid engineering, molecular cloning, plasmid prep, and sequencing
- Conduct AAV vector production using both adherent and suspension systems
- Optimize the process to increase production yield and improve production quality when needed
- Conduct AAV purification using both affinity chromatography and gradient ultracentrifugation
- Conduct analytical quality control testing for AAV vectors
- Produce high quality AAV vectors timely for in vitro and in vivo studies to support pipeline program development
- Closely interact with other functional groups including vector screening and process and analytical development teams
- Efficiently manage CROs and/or external collaborations
- Contribute to experimental design, data analysis, and report
- Maintain detailed and well-organized documentation with best industry practice
- Self-motivated, highly collaborative and passion to working in interdisciplinary environment
Skills and Requirements:
- A Ph.D. in molecular biology, cell biology, virology, immunology, or related discipline required
- Hands-on experience with AAV production and purification highly preferred
- Hands-on experience with AAV capsid discovery and engineering preferred
- Familiar with HEK293 cells and/or baculovirus/Sf9 system preferred
- Extensive and excellent hands-on skills and experience with molecular biology, cell culture techniques, and cell-based assays
- Proficient in cell culture, transfection, Western, ELISA, flow, ddPCR techniques
- Outstanding critical thinking and problem-solving skills
- Responsible and efficient with the lab assignments
- Can do attitude and quick and high-quality execution
- Ability to work under pressure and meet tight deadlines, where necessary
- Ability to be highly productive in a fluid, fast-paced team environment
